Overview

SRI-9439 is a potent lipophilic antifolate inhibitor developed by Southern Research Institute. It targets and inhibits human dihydrofolate reductase (hDHFR), forming a ternary complex with NADPH. The drug was initially developed against *Toxoplasma gondii* DHFR-thymidylate synthase, indicating its potential as an anti-infective agent. Its crystal structure in complex with hDHFR and NADPH has been determined at atomic resolution, providing insights into its binding characteristics and inhibitory activity.
