SRK-181 (linavonkibart) is a fully human IgG4 monoclonal antibody that selectively inhibits the activation of latent transforming growth factor-beta 1 (TGFβ1). TGFβ1 is implicated in tumor immune escape by promoting an immunosuppressive tumor microenvironment, reducing antigen presentation and impairing T-cell infiltration and cytotoxicity. By targeting the precursor (latent) form of TGFβ1 with high specificity—without significant binding to other isoforms or active TGFβ—SRK-181 aims to overcome primary resistance to immune checkpoint inhibitors such as anti-PD-(L)1 therapies. It is being developed primarily for use in combination with pembrolizumab for patients with locally advanced or metastatic solid tumors who are resistant to prior anti-PD-(L)1 therapy. Clinical trials have shown promising efficacy and a manageable safety profile across multiple tumor types including clear cell renal cell carcinoma, melanoma, urothelial carcinoma, non-small cell lung cancer, and head and neck squamous cell carcinoma[1][2][3][5][7].
