Overview

SRK-256 is a preclinical, investigational monoclonal antibody developed by Scholar Rock that selectively targets repulsive guidance molecule C (RGMc), also known as hemojuvelin (HJV). By binding to HJV, SRK-256 inhibits its interaction with bone morphogenetic proteins (BMPs)—specifically BMP2, BMP4, and BMP6—thereby blocking the BMP signaling pathway in the liver. This pathway regulates the expression of hepcidin, a key hormone controlling iron homeostasis. In conditions such as iron-restricted anemia associated with chronic inflammation, chronic kidney disease, or myelofibrosis, elevated hepcidin traps iron within cells and limits its availability for red blood cell production. By inhibiting HJV-mediated BMP signaling and reducing hepcidin levels, SRK-256 increases serum iron availability and represents a promising therapeutic approach for treating functional iron deficiency anemias[1][3][5][6][8].
