Overview

SRK-373 is a preclinical, highly selective monoclonal antibody developed by Scholar Rock for the treatment of fibrotic diseases. It functions as a context-dependent inhibitor of latent transforming growth factor beta 1 (TGFβ1) activation, specifically targeting proTGFβ1 associated with the extracellular matrix via latent TGFβ-binding proteins LTBP1 and LTBP3. This selectivity allows SRK-373 to inhibit disease-driving TGFβ1 signaling in fibrotic tissues while sparing immune cell-associated TGFβ1, thus maintaining normal immunosuppressive function and minimizing adverse effects. The approach aims to suppress pro-fibrotic signaling in multiple organs without affecting other isoforms (TGFβ2 or TGFβ3), which have historically complicated non-selective anti-TGFβ therapies. Preclinical studies suggest that this strategy could offer a novel and safer way to treat fibrosis across various organ systems[1][2][4][5][7].
