srKD033 is a murine cross-reactive surrogate of the human immunocytokine KD033, developed by Kadmon for preclinical cancer immunotherapy research. It is a recombinant fusion protein consisting of a high-affinity antibody targeting programmed death-ligand 1 (PD-L1) fused to the sushi-domain of the human IL-15/IL-15 receptor alpha (IL-15/IL-15Rα) complex. The molecule is engineered with a LALA (L234A/L235A) modification in the Fc region to minimize antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity. srKD033 functions by blocking the inhibitory PD-1/PD-L1 checkpoint while simultaneously delivering IL-15 directly to the tumor microenvironment (TME). This localized delivery facilitates the trans-presentation of IL-15 to NK and CD8+ T cells expressing the IL-15Rβγ receptor, leading to the expansion and activation of cytotoxic effector cells. Preclinical studies in multiple syngeneic murine models have demonstrated that srKD033 induces robust antitumor activity and durable immune memory with reduced systemic toxicity compared to non-targeted IL-15 therapies.
