SROS-201 is a preclinical nanoparticle-based cancer immunotherapy developed by Saros Therapeutics that targets the Stimulator of Interferon Genes (STING) pathway. The drug utilizes a proprietary STING agonist-manganese nanoparticle (SMP) technology, originally developed at the University of Michigan, which combines a STING agonist with manganese ions to significantly amplify immune activation (up to 77-fold compared to agonists alone). These components are formulated into nano-sized crystals and encapsulated within a lipid layer to enable systemic (intravenous) delivery, addressing the historical challenge of STING agonists requiring intratumoral injection due to poor stability and systemic toxicity. SROS-201 is designed to target both tumor tissues and key innate immune cells, such as dendritic cells and macrophages, to initiate a robust anti-tumor immune response and turn "cold" tumors "hot," potentially enhancing the efficacy of existing immunotherapies like checkpoint inhibitors.
