SRT-015 is a highly optimized, second-generation, liver-selective small molecule inhibitor of apoptosis signal-regulating kinase 1 (ASK1), also known as MAP kinase kinase kinase 5. It is being developed primarily for the treatment of severe acute alcoholic hepatitis (AH), non-alcoholic steatohepatitis (NASH), and other orphan liver diseases. SRT-015 was designed to overcome the limitations of first-generation ASK1 inhibitors such as selonsertib, offering improved safety and a liver-preferential distribution profile. The drug demonstrates anti-inflammatory, anti-fibrotic, and anti-apoptotic effects in preclinical models by inhibiting ASK1-mediated signaling pathways involved in hepatic inflammation and fibrosis. Clinical studies have shown that SRT-015 is safe and well-tolerated in healthy volunteers at various dose levels. The developer has also explored injectable formulations for acute-on-chronic liver disease (ACLF). Development has been discontinued for amyotrophic lateral sclerosis (ALS), retinal disorders, and skin disorders.
