SS-001

SS-001 is a topical ointment formulation of sildenafil, a phosphodiesterase type 5 (PDE5) inhibitor, developed by Suzhou Pairui Biotechnology for the prevention and treatment of chemotherapy-induced hand-foot syndrome (HFS). HFS, also known as palmar-plantar erythrodysesthesia, is a common dose-limiting toxicity associated with chemotherapeutic agents like capecitabine and 5-fluorouracil. The mechanism of SS-001 involves the inhibition of PDE5 in the skin of the hands and feet, which increases cyclic guanosine monophosphate (cGMP) levels, leading to vasodilation and improved local microcirculation. This improved blood flow is hypothesized to facilitate the clearance of toxic chemotherapy metabolites or prevent their accumulation in the small capillaries of the extremities. SS-001 is currently undergoing Phase 2 clinical trials in China to evaluate its efficacy and safety in cancer patients receiving capecitabine-based regimens.
