SS-1 is a standardized Chinese herbal formula developed for the treatment of Sjögren’s syndrome. It is a combination of three traditional Chinese medicine (TCM) formulas: Gan-Lu-Yin, Sang-Ju-Yin, and Xue-Fu-Zhu-Yu decoction, mixed in a 2:1:1 ratio. The formula is designed to address the "yin-deficiency," "dryness-heat," and "stasis" associated with the disease in TCM by exerting anti-inflammatory, immunomodulatory, and anti-fibrotic effects. Mechanistically, SS-1 has been shown to inhibit B-cell maturation antigen (BCMA) and dampen the levels of B-cell activating factor (BAFF), which are critical in the pathogenesis of Sjögren’s syndrome. It also reduces the expression of pro-inflammatory cytokines such as IL-17A, IL-18, and IL-23, and matrix metalloproteinase-9 (MMP-9). Clinical trial results indicate that SS-1 significantly improves tear secretion and reduces the need for artificial tears in patients with Sjögren’s syndrome.
