SS-LAR (Somatostatin Analog - Long Acting Release) is a clinical trial designation for long-acting formulations of somatostatin receptor ligands, specifically octreotide LAR and lanreotide. These drugs are synthetic peptides that mimic the action of natural somatostatin but possess a significantly longer duration of action, typically allowing for once-monthly administration. They function by binding with high affinity to somatostatin receptors (SSTRs), most notably SSTR2 and SSTR5, which are frequently overexpressed on the surface of neuroendocrine tumor (NET) cells. Activation of these receptors leads to the inhibition of hormone secretion (such as serotonin, growth hormone, and gastrin) and suppresses tumor cell proliferation through various intracellular signaling pathways. SS-LARs are established standard-of-care therapies for managing carcinoid syndrome and controlling tumor growth in patients with well-differentiated gastro-entero-pancreatic neuroendocrine tumors (GEP-NETs). In the context of the Lu-PRRT-Cap trial, SS-LAR is utilized as a maintenance therapy following peptide receptor radionuclide therapy (PRRT).
