SS1-28z CAR T cells are an investigational second-generation chimeric antigen receptor (CAR) T-cell therapy targeting mesothelin, a cell-surface glycoprotein overexpressed in various solid tumors. The CAR construct incorporates the SS1 single-chain variable fragment (scFv) for antigen binding, linked to a CD28 costimulatory domain and a CD3ζ signaling endodomain. This design is engineered to provide rapid T-cell activation and potent cytolytic activity upon engagement with mesothelin-expressing tumor cells. Developed through research at institutions like the National Cancer Institute and the University of Pennsylvania, SS1-28z has been evaluated in preclinical and clinical studies for the treatment of mesothelin-positive malignancies, including mesothelioma, pancreatic cancer, and ovarian cancer. While the CD28 domain promotes robust effector function, it is often characterized by rapid expansion followed by limited long-term persistence compared to 4-1BB-based constructs.
