**ssAAV2-STC-1-FLAG** is a gene therapy product based on a single-stranded adeno-associated virus serotype 2 (ssAAV2) vector engineered to express stanniocalcin-1 (STC-1) fused to a FLAG tag. This vector is designed for sustained reduction of intraocular pressure (IOP) primarily for the treatment of glaucoma and ocular hypertension. The mechanism involves gene transfer, resulting in intraocular production of the STC-1 protein, a secreted peptide hormone with IOP-lowering and neuroprotective properties. IOP reduction is achieved via increased aqueous outflow facility and does not require activation of the prostaglandin F (FP) receptor. The gene therapy product can be administered by intracameral (anterior chamber) or subconjunctival injection, and has been shown in preclinical mouse studies to result in sustained IOP lowering for up to 6 months per injection, with effects reproducible upon repeat administration. The FLAG tag is used for detection and analysis of the transgene expression[1][2].
