SSR-126517E is a potent and selective non-peptide antagonist of the arginine vasopressin receptor 1B (AVPR1B), formerly known as the V3 receptor. Developed by Sanofi, the compound was primarily investigated for its potential in treating stress-related psychiatric disorders, including major depressive disorder and generalized anxiety disorder. By blocking AVPR1B receptors in the anterior pituitary, SSR-126517E modulates the hypothalamic-pituitary-adrenal (HPA) axis by inhibiting vasopressin-induced release of adrenocorticotropic hormone (ACTH). Preclinical studies demonstrated that the drug exhibits significant anxiolytic-like and antidepressant-like effects in animal models. However, clinical development was discontinued following the broader termination of Sanofi's vasopressin antagonist program, which also included the candidate nelivaptan.
