Overview

SSR128129 is a small molecule allosteric antagonist of the fibroblast growth factor receptor (FGFR), specifically targeting the extracellular domain of FGFR1. Developed by Sanofi, it inhibits FGF-2-induced signaling pathways that are critical for tumor cell proliferation, survival, and angiogenesis. Preclinical research has demonstrated that SSR128129 can act as a radiosensitizer in pancreatic cancer models, significantly enhancing the efficacy of radiotherapy by reducing microvessel density and overcoming FGFR1-mediated radioresistance. While it has shown promise in early-stage research for solid tumors and pancreatic carcinoma, it has primarily remained in the preclinical phase of development.
