SST4 agonist (Algonist Biotechnologies)

Algonist Biotechnologies is developing a pipeline of novel small molecule agonists targeting the somatostatin receptor subtype 4 (SST4) for the treatment of chronic and inflammatory pain. SST4 is a G protein-coupled receptor primarily expressed in the nervous system, where its activation mediates potent anti-nociceptive, anti-inflammatory, and anti-edematous effects. Unlike other somatostatin receptor subtypes (SST1, SST2, SST3, and SST5) that are heavily involved in endocrine regulation, SST4 activation does not significantly influence the release of growth hormone or other hormones, offering a therapeutic window for pain management without systemic endocrine side effects. The program is currently in preclinical development, with a focus on optimizing lead compounds for oral bioavailability and safety in various pain models.
