SSTK-214 is a first-in-class small molecule inhibitor of NAD kinase (NADK) being developed by StemSynergy for the treatment of Notch-driven cancers. Licensed from the University of Miami, SSTK-214 targets the metabolic dependency of these cancers on the NADK enzyme, which is responsible for converting NAD+ to NADP+. By inhibiting NADK, the drug reduces the levels of NADPH, a critical cofactor for antioxidant defense and lipid biosynthesis, thereby inducing oxidative stress and metabolic exhaustion in Notch-dependent tumor cells. It is currently in the preclinical stage of development and represents a novel metabolic approach to treating malignancies driven by the Notch signaling pathway.
