Overview

SSTN-465 is a first-in-class small molecule modulator developed by StemSynergy that specifically targets the Notch3 ternary complex (Type II). The Notch signaling pathway is a highly conserved cell signaling system that plays a critical role in cell fate determination, and its aberrant activation is a key driver in various malignancies. SSTN-465 is designed to modulate the formation or activity of the Notch3 transcriptional complex, thereby inhibiting oncogenic signaling in Notch-driven cancers. As of early 2026, the compound is in the IND-enabling preclinical stage of development.
