Overview

ST-401 is a brain-penetrant small molecule that acts as a mild inhibitor of microtubule (MT) assembly. Unlike traditional microtubule-targeting agents (MTAs) that disrupt mitosis, ST-401 kills glioblastoma (GBM) cells during interphase by disrupting mitochondrial energy supply. It binds to the colchicine site of alpha,beta-tubulin, inducing a unique structural stabilization that triggers mitochondrial fission through the activation of Dynamin-related protein 1 (Drp1). ST-401 has demonstrated significant in vivo antitumor activity in glioblastoma mouse models and is being developed for the treatment of brain tumors. Its active enantiomer is identified as ST-403.
