ST-617 is an investigational small molecule dithioethione being developed by Supportive Therapeutics for the attenuation of oral mucositis (OM) in patients undergoing chemoradiation therapy (CRT) for head and neck cancer. It acts as an activator of the **Nrf2 (Nuclear factor erythroid 2-related factor 2)** pathway, which regulates the expression of antioxidant proteins that protect against oxidative damage triggered by radiation and chemotherapy. By addressing oxidative stress, a critical event in oral mucositis pathogenesis, ST-617 aims to reduce the severity and duration of mucosal injury. The drug is administered as an oral suspension and has been evaluated in Phase 1b dose-escalation multicenter studies assessing safety, tolerability, pharmacokinetics, and preliminary efficacy in high-risk patients receiving cisplatin-based chemoradiation.
