ST101

ST101 is a first-in-class monoclonal antibody developed by Spica Therapeutics that targets CD163 (also known as Scavenger receptor cysteine-rich type 1 protein M130). CD163 is a scavenger receptor highly expressed on immunosuppressive tumor-associated macrophages (TAMs) within the tumor microenvironment. The antibody is designed to selectively deplete these CD163+ TAMs through antibody-dependent cell-mediated cytotoxicity (ADCC). To enhance its potency, ST101 utilizes ProBioGen's proprietary GlymaxX technology to achieve targeted afucosylation, which significantly boosts its ADCC activity by increasing affinity for FcγRIIIa on effector cells. By remodeling the tumor microenvironment from an immunosuppressive ('cold') state to an immunostimulatory ('hot') state, ST101 is intended to enhance the efficacy of immune checkpoint inhibitors and other cancer therapies. As of late 2025, the program is in the preclinical stage, advancing through IND-enabling studies for the treatment of solid tumors via intravenous administration.
