ST2825 is a **synthetic small molecule inhibitor** that specifically targets the homodimerization of Myeloid Differentiation primary response protein 88 (**MyD88**) by mimicking the structure of its BB-loop peptide domain[1][3][5]. By blocking MyD88 dimerization, ST2825 inhibits downstream signaling of **Toll-like receptors (TLRs)** and **interleukin-1 receptors (IL-1Rs)**, suppressing activation of the **NF-κB** pathway. This leads to decreased expression of pro-inflammatory cytokines and proteins, cell cycle arrest, and enhanced apoptosis in several cancer models (including hepatocellular carcinoma and pancreatic ductal adenocarcinoma)[1][4], as well as reduced neuroinflammation and inflammatory mediators in preclinical studies[5]. Preclinical investigations have implicated its potential for cancer, B-cell lymphoma, myocardial infarction, chronic inflammatory diseases, and neuroinflammation[2][5]. ST2825 is not approved for clinical use and is primarily used as a research tool.
