ST80 is a novel hydroxamate derivative that functions as a selective inhibitor of histone deacetylase 6 (HDAC6). It has demonstrated potent growth-inhibitory effects at low micromolar concentrations in various leukemia cell lines (HL60, Kasumi-1, NB-4, THP-1, K562, U937, Jurkat) and primary acute myeloid leukemia (AML) blasts. Its mechanism involves preferentially inducing α-tubulin acetylation, suggesting its antileukemic activity is mediated through HDAC6 inhibition rather than histone reacetylation.
