Overview

STA-9584 is a novel small molecule vascular disrupting agent (VDA) that functions as a tubulin-binding prodrug. It is metabolized into its active form, STA-9122, which selectively disrupts established tumor microvasculature by binding to tubulin and inhibiting its polymerization. Developed by Synta Pharmaceuticals, STA-9584 distinguishes itself from other agents in the combretastatin class by effectively targeting microvasculature in both the central and peripheral regions of solid tumors. Preclinical studies in prostate and breast cancer xenograft models have demonstrated significant tumor regression and superior growth inhibition compared to combretastatin A-4 phosphate (CA4P). Additionally, STA-9584 has shown a more favorable cardiovascular safety profile in research models compared to other vascular disrupting agents.
