Drug intelligence / Profile preview
Stabilized bivalent liposomal iron (SBLI) is an oral formulation of ferrous sulfate encapsulated within a protective liposomal membrane. This liposomal delivery system is designed to protect the bivalent iron from gastric acid and prevent direct contact with the gastrointestinal mucosa, thereby minimizing common side effects associated with conventional oral iron supplements, such as gastrointestinal irritation, nausea, and constipation. Crucially, the liposomal structure allows the bivalent iron to bypass the traditional hepcidin-mediated absorption pathway (which is often upregulated in chronic inflammatory states like heart failure). Instead, it undergoes direct absorption by enterocytes via endocytosis and phagocytosis, providing high bioavailability and sustained iron repletion. It has been clinically evaluated for the treatment of iron deficiency and anemia in patients with chronic heart failure (CHF), demonstrating superior long-term efficacy in maintaining iron balance and improving quality of life compared to standard intravenous ferric carboxymaltose [1].
