Overview

Staccato Granisetron is an investigational, inhaled formulation of the 5-HT3 receptor antagonist granisetron delivered using Alexza’s Staccato One Breath Technology to provide ultra-rapid systemic exposure while bypassing the gastrointestinal tract. The device rapidly heats and vaporizes excipient-free granisetron into an aerosol that is inhaled in a single breath, achieving plasma pharmacokinetics similar to intravenous granisetron but via a needle-free route. It is being developed primarily for the acute treatment of cyclic vomiting syndrome and other conditions characterized by severe episodic nausea and vomiting, leveraging granisetron’s established mechanism as a selective serotonin 5-HT3 receptor antagonist acting on central (area postrema/chemoreceptor trigger zone) and peripheral (vagal and splanchnic afferents in the GI tract) pathways.
