Stacofylline

Stacofylline is a trimethylxanthine derivative and potent acetylcholinesterase (AChE) inhibitor with an IC50 of 5-50 nM. It was investigated for potential antimigraine characteristics and cognitive disorders. In experimental studies, S9977 demonstrated moderate hypotensive and bradycardic effects, with decreased carotid blood flow at higher doses. Unlike dihydroergotamine, its effects appeared related to decreased arterial blood pressure rather than active vasoconstriction of arteriovenous anastomoses. The compound was also studied comparatively with tacrine for effects on psychomotor activity and passive avoidance in cognitive disorder models.
