Overview

STACT (S. Typhimurium-Attenuated Cancer Therapy) is a patent-protected, microbial-based immunotherapeutic technology platform developed by Actym Therapeutics for treating solid tumors. [3-13, 3-14] The platform utilizes a genetically engineered, attenuated strain of *Salmonella Typhimurium* as a vehicle that can be administered systemically. [1, 2] In preclinical studies, STACT was shown to specifically enrich in solid tumors rather than healthy tissue. [3-15, 3-16] Once inside the tumor, the bacteria deliver multiple immuno-modulatory payloads directly to tumor-resident immune cells, particularly phagocytic antigen-presenting cells (APCs), activating both innate and adaptive immunity. [2, 3-15, 3-16, 6] This approach enables the local delivery of potent therapeutic combinations that would be too toxic for systemic administration via conventional methods. [1, 3-17] The lead clinical candidate from this platform is ACTM-838, which delivers an engineered IL-15plex and a STING agonist. [2, 3]
