Overview

STACT-TREX1 is a first-in-class, systemically administered microbial-based immunotherapy developed by Actym Therapeutics using its proprietary STACT (Salmonella typhimurium (Attenuated) + Cancer Therapy) platform. The therapy consists of a genetically engineered, attenuated strain of Salmonella typhimurium designed to selectively accumulate in the tumor microenvironment. Once localized, it delivers inhibitory microRNA (RNAi) to the myeloid compartment to silence TREX1, a 3′ exonuclease that acts as an immune checkpoint by degrading cytosolic DNA and preventing the activation of the cGAS-STING pathway. By inhibiting TREX1, STACT-TREX1 promotes STING-mediated immune activation, leading to potent anti-tumor activity. Preclinical studies have demonstrated its efficacy as a monotherapy, resulting in significant tumor growth inhibition and complete regressions in models of colon carcinoma and melanoma.
