Overview

SyntheX Labs is developing a small-molecule molecular glue targeting STAG1 for the treatment of cancer. Discovered using the company's proprietary ToRNeDO platform, this program leverages the concept of synthetic lethality. STAG1 is a paralog of STAG2, both of which are components of the cohesin complex. In many cancers, STAG2 is mutated or lost; research indicates that these STAG2-deficient cells become uniquely dependent on STAG1 for survival. By promoting the degradation or functional modulation of STAG1 through a molecular glue mechanism, the drug aims to selectively kill STAG2-deficient tumor cells while sparing healthy tissue. The program is currently in the screening and discovery phase.
