Drug intelligence / Profile preview
STAM2 mRNA-LNP is an mRNA-based therapeutic delivered via lipid nanoparticles (LNPs) that encodes signal transducing adaptor molecule 2 (STAM2). Developed by researchers at Wuhan University, it is designed to target osteoclast precursors to prevent and treat weightlessness-induced bone loss and other mechanical skeletal disorders. STAM2 is a component of the endosomal sorting complex required for transport (ESCRT) pathway. By restoring STAM2 expression, the therapy promotes the degradation of serine/threonine kinase 38 (STK38), a Hippo pathway kinase. This degradation prevents the phosphorylation-dependent cytoplasmic retention of TAZ, restoring its nuclear translocation and maintaining redox homeostasis by reducing reactive oxygen species (ROS) accumulation, which ultimately suppresses excessive osteoclast differentiation and bone resorption.
