This chemotherapy regimen refers to the specific multi-agent backbone used as the standard-of-care control arm in the Phase II BERNIE trial (NCT00702234), sponsored by Hoffmann-La Roche. The regimen was designed for pediatric and adolescent patients with metastatic rhabdomyosarcoma (RMS) or non-rhabdomyosarcoma soft tissue sarcoma (NRSTS). It consists of an intensive induction phase followed by a maintenance phase. Induction therapy involves either the IVADo regimen (ifosfamide, vincristine, dactinomycin, and doxorubicin) or the IVA regimen (ifosfamide, vincristine, and dactinomycin). This is followed by a maintenance phase consisting of vinorelbine and cyclophosphamide. The combination targets cancer cells through multiple cytotoxic pathways, including DNA cross-linking, microtubule disruption, and inhibition of DNA replication and transcription.
