Overview

Staphylococcal enterotoxin B (SEB) plasmid DNA is a genetic construct that carries the gene encoding for Staphylococcal enterotoxin B, a potent bacterial superantigen derived from *Staphylococcus aureus*. This plasmid DNA is being explored as a therapeutic DNA vaccine, primarily in the context of cancer immunotherapy. Upon administration, the plasmid facilitates the expression of SEB within host cells. The expressed SEB then acts as a superantigen, capable of activating a significant proportion of T cells by simultaneously binding to the variable beta chain of the T cell receptor (Vβ-TCR) and major histocompatibility complex class II (MHC II) molecules on antigen-presenting cells. This robust T cell activation leads to the release of pro-inflammatory cytokines, such as interferon-gamma (IFN-γ), interleukin-2 (IL-2), and tumor necrosis factor-alpha (TNF-α), which collectively contribute to an anti-tumor immune response, including tumor necrosis and reduced tumor growth.
