Overview

The Staphylococcus aureus 4-Antigen Vaccine (SA4Ag) is an investigational vaccine developed to prevent invasive infections caused by Staphylococcus aureus, particularly in adults undergoing elective surgical procedures. The vaccine contains four antigens targeting three key virulence mechanisms of S. aureus that are critical for the initiation and maintenance of infection. These antigens include capsular polysaccharide conjugates of serotypes 5 (CP5) and 8 (CP8), each linked to CRM197 (a nontoxic mutant diphtheria toxin), a recombinant form of clumping factor A (ClfA Y338A mutant), and a recombinant non-lipidated manganese transporter subunit C (MntC/rP305A). The mechanism of action involves eliciting functional immune responses with robust opsonophagocytic activity against S. aureus by blocking bacterial adhesion, nutrient acquisition, and evasion of neutrophil-mediated killing. Clinical studies have shown that the vaccine is generally safe and induces strong antibody responses; however, efficacy in preventing infection during surgery has not been demonstrated conclusively[1][2][3][6][8].
