Overview

The Staphylococcus aureus type 336 vaccine is a conjugate vaccine candidate designed to provide protection against S. aureus strains that do not express the common serotype 5 or 8 capsular polysaccharides. It utilizes the type 336 capsular polysaccharide (336PS) as the primary antigen, which is chemically conjugated to a carrier protein—most commonly recombinant Pseudomonas aeruginosa exotoxin A (rEPA)—to induce a T-cell dependent immune response and produce opsonophagocytic antibodies. Developed by Nabi Biopharmaceuticals (later acquired by GSK), the type 336 antigen was integrated into multivalent formulations, including a trivalent vaccine (targeting types 5, 8, and 336) and a pentavalent vaccine known as PentaStaph (or Nabi-StaphVAX+), which also included antigens against Staphylococcus epidermidis. These vaccines were primarily investigated for the prevention of S. aureus bacteremia in high-risk individuals, such as hemodialysis patients, but development was discontinued following the failure of related clinical trials and corporate acquisitions.
