The staphylococcus aureus type 5 capsular polysaccharide conjugate vaccine is designed to prevent infections caused by *Staphylococcus aureus* by targeting its type 5 capsular polysaccharide. This polysaccharide is a crucial virulence factor that enables the bacterium to evade the host's immune response. By conjugating the type 5 capsular polysaccharide to a carrier protein, such as CRM197 (a nontoxic mutant of diphtheria toxin) or *Pseudomonas aeruginosa* exoprotein A (rEPA), the vaccine significantly enhances immunogenicity. This conjugation leads to a robust immune response, primarily characterized by the production of opsonophagocytic antibodies. These antibodies bind to the bacterial capsule, facilitating opsonization and subsequent phagocytic clearance of *S. aureus* by immune cells, thereby providing protection against various staphylococcal infections. While often developed as part of bivalent vaccines that also include type 8 capsular polysaccharide, the type 5 component is a key antigen in these formulations. It has been investigated for preventing bacteremia in high-risk populations, though some specific bivalent formulations have faced challenges in clinical trials.
