STAT-6 siRNA is a small interfering RNA (siRNA) designed to specifically silence the expression of the signal transducer and activator of transcription 6 (STAT6) gene. STAT6 is a transcription factor involved in cytokine signaling pathways, particularly those mediated by interleukin-4 (IL-4), and plays a key role in immune responses as well as cancer cell proliferation. By targeting and degrading STAT6 mRNA via the RNA interference pathway, STAT-6 siRNA inhibits both mRNA and protein levels of STAT6 within cells. This results in reduced cell proliferation and increased apoptosis in various cancer cell lines that overexpress STAT6, such as colorectal, breast, lung adenocarcinoma cells, among others[4][5][1]. Preclinical studies have also explored its use for allergic diseases like asthma by delivering it locally to respiratory tract cells using liposomes or aerosolized formulations[2]. Delivery systems under investigation include gelatin nanocarriers for enhanced stability and cellular uptake[1].
