Overview

STAT3 antisense oligonucleotides (ASOs) are a class of therapeutic agents designed to selectively inhibit the expression of Signal Transducer and Activator of Transcription 3 (STAT3), a transcription factor that is frequently overactivated in a wide range of cancers. STAT3 plays a critical role in promoting tumor cell proliferation, survival, and metastasis, while also contributing to an immunosuppressive tumor microenvironment by regulating the production of pro-tumorigenic cytokines such as IL-6 and IL-1β. These ASOs function by binding to STAT3 mRNA through complementary base pairing, which triggers RNase H-mediated degradation of the transcript and a subsequent reduction in STAT3 protein levels. This approach aims to overcome the challenges of targeting STAT3 with traditional small molecules. The most prominent candidate in this class, danvatirsen (also known as AZD9150 or ISIS 481464), was developed by Ionis Pharmaceuticals in collaboration with AstraZeneca and has been investigated in Phase 2 clinical trials for conditions such as diffuse large B-cell lymphoma (DLBCL) and non-small cell lung cancer (NSCLC).
