STAT3 inhibitor (topical)

A topical STAT3 inhibitor refers to any formulation of a therapeutic agent designed for local application to the skin that inhibits the Signal Transducer and Activator of Transcription 3 (STAT3) protein. STAT3 is a key transcription factor downstream of various cytokine receptors (such as IL-6 and IL-22) and Janus kinases (JAKs). In inflammatory skin conditions like dermatitis and psoriasis, STAT3 activation in keratinocytes drives epidermal hyperplasia, barrier dysfunction, and the expression of pro-inflammatory cytokines and chemokines (such as IL-6 and KRT17). Topical administration of STAT3 inhibitors aims to suppress this localized inflammatory cascade and restore skin barrier integrity while minimizing systemic side effects.
