Overview

**STAT3 siRNA** is a synthetic double-stranded small interfering RNA specifically designed to target and silence the expression of the STAT3 gene by RNA interference. STAT3 (Signal Transducer and Activator of Transcription 3) is a transcription factor frequently overactivated in various cancers, including chronic myeloid leukemia (CML), glioblastoma multiforme (GBM), and certain lymphomas, where it promotes tumor cell proliferation, survival, resistance to apoptosis, and contributes to chemotherapeutic resistance. STAT3 siRNA binds to STAT3 mRNA, promoting its degradation and inhibiting translation, thereby reducing STAT3 protein levels and blocking its transcriptional activity[1][2][3]. This leads to tumor cell apoptosis, decreased cell survival, and metabolic reprogramming (switching cancer cells from glycolysis to mitochondrial respiration), restoring sensitivity to treatments such as tyrosine kinase inhibitors like nilotinib[1]. In preclinical studies, lipopolyplex nanoparticle delivery of STAT3 siRNA effectively suppressed tumor growth and improved survival in glioma models[2]. Novel formulations, such as CpG-STAT3 siRNA, combine CpG oligonucleotides (to stimulate immune responses via TLR9) with STAT3 siRNA, achieving both gene silencing and activation of systemic antitumor immunity[3][4].
