Overview

StatC (also known as TZLS-101) is a small molecule inhibitor of the Signal Transducer and Activator of Transcription 3 (STAT3) protein, a transcription factor that plays a pivotal role in oncogenesis and immune modulation. STAT3 is frequently hyperactivated in various malignancies, where it drives the expression of genes associated with cell survival, proliferation, and metastasis. StatC works by binding to the SH2 domain of STAT3, thereby inhibiting its phosphorylation and dimerization, which are essential steps for its translocation to the nucleus and subsequent DNA binding. Originally developed at Cardiff University and licensed by Tiziana Life Sciences, StatC was primarily investigated for the treatment of solid tumors, including breast and colorectal cancers. Although it showed promise in preclinical models for its ability to induce apoptosis in cancer cells and inhibit tumor growth, its clinical development has been less prominent in recent years compared to other pipeline candidates.
