Overview

STATegics' thrombopoietin (TPO) allomimetics are a series of preclinical small molecule allosteric modulators targeting the thrombopoietin receptor (also known as MPL). Developed as second-generation TPO mimetics, these compounds are designed to treat various forms of thrombocytopenia, including chemotherapy-induced thrombocytopenia (CIT) and immune thrombocytopenic purpura (ITP). Unlike traditional orthosteric agonists, these allosteric modulators aim to provide a more controlled activation of the TPO receptor, which may offer safety advantages in the context of bone marrow malignancies and leukemia where excessive TPO signaling can be detrimental. The program focuses on identifying orally bioavailable candidates that can stimulate megakaryopoiesis and platelet production by modulating the receptor's signaling pathways from an allosteric site.
