STATegics' thrombopoietin (TPO) allomimetics are a series of preclinical small molecule allosteric modulators targeting the thrombopoietin receptor (also known as MPL). Developed as second-generation TPO mimetics, these compounds are designed to treat various forms of thrombocytopenia, including chemotherapy-induced thrombocytopenia (CIT) and immune thrombocytopenic purpura (ITP). Unlike traditional orthosteric agonists, these allosteric modulators aim to provide a more controlled activation of the TPO receptor, which may offer safety advantages in the context of bone marrow malignancies and leukemia where excessive TPO signaling can be detrimental.
