Overview

Stattic is a small molecule inhibitor that selectively targets the SH2 domain of Signal Transducer and Activator of Transcription 3 (STAT3), preventing its dimerization and activation regardless of phosphorylation state. By blocking STAT3 activity—including phosphorylation at Y705 and S727—Stattic inhibits downstream signaling events such as nuclear translocation and transcriptional activation. This leads to reduced cell viability in STAT3-dependent tumor cells and can induce apoptosis in certain cancer cell lines. Stattic has been used primarily as a research tool to study the role of STAT3 in cancer biology[1][2][4][5].
