Overview

Stavudine is a synthetic nucleoside analog of thymidine and belongs to the class of nucleoside reverse transcriptase inhibitors (NRTIs). It is used in combination with other antiretroviral agents for the treatment of human immunodeficiency virus (HIV) infection. Stavudine works by being phosphorylated within cells to its active triphosphate form, which inhibits HIV-1 reverse transcriptase both by competing with natural substrates and by incorporating into viral DNA, causing chain termination due to the absence of a 3'-OH group. This action prevents further elongation of viral DNA and thus inhibits replication of HIV[2][3][6][7]. Stavudine was developed as an antiviral agent specifically targeting HIV/AIDS.
