STC-1010 is a novel, off-the-shelf allogenic cancer vaccine developed as an immunotherapy for advanced or metastatic colorectal cancer. It is based on a two-step modification of established colorectal cancer cell lines to express native tumor-specific antigens (TSA) and tumor-associated antigens (TAA), with increased immunogenicity achieved through haptenization and stress induction (irradiation, heat shock, chemotherapy). The vaccine aims to stimulate the immune system by activating dendritic cells and priming CD8+ T cells, leading to robust anti-tumor responses including apoptosis of colorectal cancer cells. Preclinical studies demonstrated efficacy in both “cold” (pMMR/MSS) and “hot” (MSI-H/dMMR) tumor models, showing reduced tumor volume, increased T-cell infiltration, M1 macrophage polarization, and improved survival. In clinical development, STC-1010 is administered alongside low-dose immunostimulants (cyclophosphamide and GM-CSF) with or without standard-of-care chemotherapy regimens such as FOLFOX or FOLFIRI. The drug addresses the unmet need for effective therapies in microsatellite stable colorectal cancers that are typically unresponsive to current checkpoint inhibitors[1][3][5][7][8][9].
