Stealth-Body Fc-MMAE ADC is an experimental antibody-drug conjugate (ADC) developed by CrossPoint Therapeutics. It utilizes the proprietary Stealth-Body (SB) platform, which incorporates specific mutations in the Fc region of the antibody to eliminate interactions with Fcγ receptors (FcγRs) and C1q. This fully Fc-silent design is intended to reduce off-target toxicities, such as those caused by the non-specific uptake of ADCs by FcγR-expressing cells, thereby improving the therapeutic index. The ADC is conjugated with the cytotoxic payload monomethyl auristatin E (MMAE), a potent microtubule inhibitor. Preclinical data presented at AACR 2026 demonstrated that the SB Fc-MMAE ADC maintains strong anti-tumor efficacy and a favorable pharmacokinetic profile, with a minimum efficacious dose of 0.6 mg/kg in xenograft models. PK studies in mice and toxicology studies in non-human primates are currently ongoing to support clinical development.
