STEAP2-CAR T cell is an investigational, multi-pronged, all-in-one chimeric antigen receptor (CAR) T cell therapy designed for the treatment of metastatic castration-resistant prostate cancer (mCRPC) and other solid tumors. This advanced CAR T cell platform incorporates triple-armoring and dual-targeting functionalities to enhance anti-tumor activity, reduce on-target/off-tumor toxicities, and overcome the immunosuppressive tumor microenvironment. It targets the prostate cancer-associated antigen STEAP2 via the CAR, while also secreting an αPSMA×CD28 bispecific molecule upon STEAP2 engagement to activate bystander T cells against PSMA-positive variants. Furthermore, the CAR T cells are armored with a dominant-negative TGF-β receptor II (dnTGFβRII) to counteract TGF-β-mediated immunosuppression. This design aims to improve safety and efficacy compared to earlier CAR T cell approaches that faced challenges with toxicity and antigen heterogeneity.
