Overview

Stenoparib is an **orally-available small molecule dual inhibitor** targeting **poly(ADP-ribose) polymerases 1 and 2 (PARP1/2)** and **tankyrase 1 and 2**, enzymes centrally involved in DNA repair and the Wnt/β-catenin signaling pathway[1][4][6][7]. By blocking both pathways, stenoparib interrupts cancer cell proliferation and DNA repair, offering a differentiated mechanism among PARP inhibitors and unique activity against tumors, particularly ovarian cancer including platinum-resistant and platinum-ineligible cases[1][4][5][6][7]. It is currently in Phase 2 trials for advanced ovarian cancer and previously demonstrated clinical benefit and favorable tolerability. Developed originally by Eisai Co. Ltd., stenoparib is now under exclusive rights for further development by Allarity Therapeutics[1][4][5][7]. Besides its antineoplastic action, stenoparib also demonstrates antiviral effects against SARS-CoV-2 and other coronaviruses *in vitro*[10].
