STI-1492 is an allogeneic, off-the-shelf dimeric antigen receptor T (DAR-T) cell therapy developed by Sorrento Therapeutics for the treatment of relapsed or refractory multiple myeloma. The therapy uses healthy donor-derived T cells that are genetically engineered using non-viral gene editing to express a dimeric Fab structure of an anti-CD38 antibody on their surface, along with 4-1BB and CD3 zeta intracellular signaling domains for T cell activation. The TCR alpha gene is knocked out to prevent graft-versus-host disease (GvHD), allowing safe administration of these allogeneic cells. Upon intravenous infusion, these engineered T cells selectively recognize and kill tumor cells expressing high levels of CD38, a marker commonly found on multiple myeloma cells. STI-1492 is currently in Phase 1 clinical trials for relapsed or refractory multiple myeloma[1][2][5][6].
