Overview

STI-6643 is a fully human IgG4 monoclonal antibody that targets CD47, a cell surface glycoprotein involved in immune evasion by cancer cells. By binding to CD47, STI-6643 blocks its interaction with signal regulatory protein alpha (SIRPα) on myeloid cells, thereby inhibiting the "don't eat me" signal and promoting macrophage-mediated phagocytosis of tumor cells. The antibody has been affinity-engineered to reduce binding to normal cells such as red blood cells and lymphocytes, minimizing off-tumor toxicity while maintaining potent anti-tumor activity. Preclinical studies have shown that STI-6643 preserves T cell functionality and demonstrates favorable safety margins compared to other anti-CD47 antibodies. It is being developed primarily for the treatment of solid tumors and hematologic malignancies.
