Overview

STI-9167 is a fully human, Fc-engineered monoclonal neutralizing antibody targeting the SARS-CoV-2 spike protein receptor-binding domain, developed by Sorrento Therapeutics in collaboration with the Icahn School of Medicine at Mount Sinai as a broad-spectrum antiviral candidate for the treatment of COVID-19.[3][5] It was selected from large panels of anti–spike antibodies for its high-affinity binding and potent neutralization of all tested variants of concern, including Omicron BA.1, BA.1+R346K, and BA.2, and is engineered with reduced Fc gamma receptor interactions (LALA-type modifications) to minimize antibody-dependent enhancement while preserving stability.[3][5][10] In preclinical models, STI-9167 administered intravenously or intranasally protected susceptible mice from Omicron challenge by preventing weight loss and reducing lung viral titers to undetectable levels, and is being advanced clinically primarily as an intravenous formulation (COVISHIELD) with planned or ongoing Phase 1 and subsequent Phase 2/3 trials in COVID-19 patients.[1][3][4][8][9]
